Aborto e placentite micótica por Aspergillus fumigatus em uma égua Ciência Rural

É descrito um caso de placentite micótica e aborto em uma égua com 7 meses de gestação. Alterações macroscópicas incluíam subdesenvolvido, áreas brancas na placenta e espessamento da membrana cório-alantóide. No pulmão fetal havia duas nodulações com 1 cm de diâmetro. Microscopicamente as lesões placentárias eram predominantemente necrótico-purulentas com focos de mineralização na placenta e trombose com inflamação purulenta no pulmão do feto. Hifas fúngicas septadas e ramificadas estavam presentes na placenta e também nos nódulos pulmonares. Aspergillus fumigatus foi isolado das lesões placentárias. O diagnóstico baseou-se nos achados histopatológicos e no isolamento do agente.

Aspergilose em avestruz (Struthio camelus) no Brasil Ciência Rural

Criatórios de avestruz têm se tornado comuns no Brasil, mas por se tratar de espécie exótica, as informações sobre as doenças desses animais no nosso ambiente são escassas. Um avestruz de cinco meses de idade apresentou tosse, anorexia, perda de peso, dispnéia discreta, temperatura corporal normal e morreu oito dias após o início dos sinais clínicos. Macroscopicamente foram observados nódulos múltiplos no pulmão e nos sacos aéreos. Histolopatogicamente, observou-se pneumonia granulomatosa e necrosante multifocal com hifas ramificadas e septadas intralesionais com características morfológicas de Aspergillus sp. Fungal infections of the central nervous system (CNS) are almost always a surprising finding. Their presentation is usually subtle, often without any diagnostic characteristics, and they are frequently mistaken for pyogenic abscesses, or brain tumors. Aspergillosis of the central nervous system is an uncommon infection, mainly occurring in immunocompromised patients. It may present in several forms, including meningitis, mycotic aneurysms, infarcts and a tumoral form. Cloning and expression of an endoglucanase gene from the thermotolerant fungus Aspergillus fumigatus DBiNU-1 in Kluyveromyces lactis BJM

Abstract An intronless endoglucanase from thermotolerant Aspergillus fumigatus DBINU-1 was cloned, characterized and expressed in the yeast Kluyveromyces lactis. The full-length open reading frame of the endoglucanase gene from A. fumigatus DBiNU-1, designated Cel7, was 1383 nucleotides in length and encoded a protein of 460 amino acid residues. The predicted molecular weight and the isoelectric point of the A. fumigatus Cel7 gene product were 48.19 kDa and 5.03, respectively. Contribution of gliotoxin to aspergillosis OAK

Aspergillus species (spp.) are medically important pathogens that cause aspergillosis, and A. fumigatus is the most prevalent causative agent of the disease. Aspergillus spp. produce various secondary metabolites; however, the role of these metabolites in aspergillosis remains unclear. Gliotoxin is a major secondary metabolite produced by A. fumigatus. Several recent studies have reported that gliotoxin contributes to the virulence of A. fumigatus. Here I review studies related to gliotoxin and its contribution to the infection caused by A. fumigatus.

Decolourization of textile dye effluent by non-viable biomass of Aspergillus fumigatus BABT

The aim of this work was to study the decolourization of textile dye effluent by non-viable biomass of Aspergillus fumigates. The dried non-viable fungal biomass exhibited maximum dye removal at pH 7.0 with temperature of 30ºC and 3 g/l (w/v) biomass concentration, after 24 h contact time. The results showed that the non-viable biomass possessed high efficiency for dye removal from textile effluent.

Elastin increases biofilm and extracellular matrix production of Aspergillus fumigatus BJM

Abstract Aspergillus fumigatus is an opportunistic saprobe fungus that accounts for 90% of cases of pulmonary aspergillosis in immunosuppressed patients and is known for its angiotropism. When it reaches the respiratory tract, A. fumigatus interacts with structural components and blood vessels of the lungs, such as elastin. To understand the effect of this structural component, we examined the effect of elastin on the production and development of the biofilm of A. fumigatus. In vitro antifungal susceptibility of clinical and environmental isolates of Aspergillus fumigatus and Aspergillus flavus in Brazil BJID

ABSTRACT The in vitro susceptibility of 105 clinical and environmental strains of Aspergillus fumigatus and Aspergillus flavus to antifungal drugs, such as amphotericin B, azoles, and echinocandins was evaluated by the broth microdilution method proposed by the European Committee on Antimicrobial Susceptibility Testing (EUCAST). Following the EUCAST-proposed breakpoints, 20% and 25% of the clinical and environmental isolates of A. fumigatus, respectively, were found to be resistant to itraconazole (Minimal Inhibitory Concentration, MIC > 2.0 mg/L). Aspergillus is a ubiquitous fungus which can cause a variety ofclinical syndromes. This fungus has emerged as agent of systemic infections and has therefore gained considerable public health importance. This paper describes two cases of invasive aspergillosis caused by A. fumigatus in immuno-suppressed patients and underscores the importance of early identification of Aspergillus infection associated with systemic lupus erythematosus and cardiac postoperative complications.

Enzyme expression in different fermentation systems yields important information for improving our understanding of enzymatic production induction. Comparative studies between solid-state fermentation (SSF) using agro-industrial waste wheat bran and submerged fermentation (SmF) using synthetic media were carried out to determinate the best parameters for peptidase production by the fungus Aspergillus fumigatus Fresen. Purification and characterization of a low molecular weight xylanase from solid-state cultures of Aspergillus fumigatus Fresenius Rev. Microbiol.

A xylan-degrading enzyme (xylanase II) was purified to apparent homogeneity from solid-state cultures of Aspergillus fumigatus Fresenius. The molecular weight of xylanase II was found to be 19 and 8.5 kDa, as estimated by SDS-PAGE and gel filtration on FPLC, respectively. The purified enzyme was most active at 55 °C and pH 5.5. It was specific to xylan. The apparent Km and Vmax values on soluble and insoluble xylans from oat spelt and birchwood showed that xylanase II was most active on soluble birchwood xylan. Purification and characterization of an alpha-galactosidase from Aspergillus fumigatus BABT

Aspergillus fumigatus secreted invertase (beta-fructofuranosidase) and alpha-galactosidase enzymatic activities able to hydrolyzing raffinose oligosaccharides (RO). alpha-Galactosidase was induced by galactose, melibiose and raffinose, but galactose was the most efficient inducer. It was purified by gel filtration and two ion exchange chromatographies and showed Mw of 54.7 kDa. The purified enzyme showed maximal activity against p-nitrophenyl-alpha-D-galactopyranoside (pNPGal) at pH 4.5-5.5 and 55 °C, and retained about 80% of the original activity after incubation for 90 minutes at 50ºC. The KM for pNPGal was 0.3 mM. Melibiose was hydrolyzed by the enzyme but raffinose was very poor substrate.

The potential of an Aspergillus fumigatus Brazilian strain to produce antimicrobial secondary metabolites BJM

During the course of screening for biologically leading active natural products from fungi, a strain of Aspergillus fumigatus was assessed for antimicrobial activity production. Four parameters consisting of medium culture, pH, temperature and time of fermentation were investigated. The extracts from the fungus cultures displayed different TLC profiles and produced, at least, one inhibition zone in the bioautography assays. Overlapping inhibition zones in the range of Rf 0.09 to 0.65 against Kocuria rhizophila were detected in the bioautography assay of the chloroform extract obtained from Jackson's medium culture, initial pH of 6.0 and incubation at 40ºC for 144 hours.
